Diluting All Four Brands of Kefir

After being in the incubator all night, the four different brands of kefir showed that they do contain
bacteria!! That was a good sign because we were doubtful that there would not be enough bacteria  
present to incubate. Since there were enough, we decided to dilute each of the brands of kefir.
We plated the second and third dilutions for incubation.


Dilution of Fresh Made Kefir:

1. Obtain 3 test tubes. Label them by their dilution factor (101, 102, or 103). 

2. Add 1.8 mL of Lysogenic Broth (LB) to 101, 102, and 103.


3. Extract 200 μL of Fresh Made kefir to 101. Shake. 


4. Extract 200 μL of 101 and add it to 102. Shake. 


5. Extract 200 μL of 102 and add it to 103. Shake. 


6. Extract 100 μL of 102. Place in in the center of the respective plate. 


7. Use the L shape spreader while rotating the plate in a circular motion to spread the solution.

Put the lid back on the plate once all of the liquid is absorbed into the media. 

8. Extract 100 μL of 103. Place in in the center of the respective plate. 


9. Use the L shape spreader while rotating the plate in a circular motion to spread the solution.
Put the lid back on the plate once all of the liquid is absorbed into the media. 

10. Incubate both plates at 37.0 degrees Celsius. Observe the bacterial growth after the plates
remain in the incubator overnight.
